Clonal evolution and KRAS-MET coamplification during secondary resistance to EGFR-targeted therapy in metastatic colorectal cancer

چکیده
Representative data from KRAS (A) and MET (B) relative gene quantification (RQ) in the three metastatic sites (liver, retroperitoneum and ureteral lesion) using TaqMan Copy Number Assay. The number of copies of the KRAS and MET genes is determined by relative quantification using the comparative CT (??CT) method. MAD1L1 and IL22 genes, respectively on chromosome 7 and 12, were used as reference genes to exclude a potential polysomy.

Resistance to anti-EGFR therapy in colorectal cancer: from heterogeneity to convergent evolution.
UNLABELLED The EGFR-targeted antibodies cetuximab and panitumumab are used to treat metastatic colorectal cancers. Mutations in KRAS, NRAS, and BRAF and amplification of ERBB2 and MET drive primary (de novo) resistance to anti-EGFR treatment. Recently, the emergence of alterations in the same genes was detected in patients who responded to EGFR blockade and then relapsed. These results illumina...
متن کاملAmplification of the MET receptor drives resistance to anti-EGFR therapies in colorectal cancer.
EGF receptor (EGFR)-targeted monoclonal antibodies are effective in a subset of metastatic colorectal cancers. Inevitably, all patients develop resistance, which occurs through emergence of KRAS mutations in approximately 50% of the cases. We show that amplification of the MET proto-oncogene is associated with acquired resistance in tumors that do not develop KRAS mutations during anti-EGFR the...
متن کاملBiomarkers predicting resistance to epidermal growth factor receptor-targeted therapy in metastatic colorectal cancer with wild-type KRAS
EGFR pathway is an important therapeutic target in human tumors, including metastatic colorectal cancer (mCRC). The advent of EGFR-targeted monoclonal antibodies panitumumab and cetuximab has generated promise for the treatment of mCRC and has largely improved patients' progression-free survival (PFS) and overall survival (OS). However, treatment with anti-EGFR monoclonal antibodies is only eff...
متن کاملCorrection: Molecular Heterogeneity and Receptor Coamplification Drive Resistance to Targeted Therapy in MET-Amplified Esophagogastric Cancer.
UNLABELLED MET inhibition is effective in some patients with MET-amplified esophagogastric cancer (EGC), but understanding acquired and de novo resistance mechanisms will be critical to improving therapy. We identified KRAS mutation as a novel cause of acquired resistance in a patient after a 2-year response to a MET inhibitor. We also observed that 40% to 50% of patients with MET-amplified EGC...
متن کاملارزیابی فراوانی جهشهای ژن KRAS در بیماران ایرانی مبتلا به سرطان کولورکتال
Background: Kirsten rat sarcoma (KRAS) gene is a target of genetic alterations which are diagnostic and prognostic biomarkers in patients with metastatic colorectal cancer who are treated with monoclonal anti-EGFR antibodies such as cetuximab and panitumumab. KRAS mutations are seen in 35-42% of patients with colorectal cancer. The high frequency of these mutations in colorectal cancer represen...
